Sequencing of RNA viruses infecting an indigenous potato (Solanum phureja) to support future seed certification programs in Colombia
Resumen
S. phureja cv. criolla is a diploid potato species than has received increasing attention in Colombia as a potential exotic export product due to its excellent organoleptic properties; high levels of vitamins A, B, C, niacine and thiamine; short production cycle and reduced tuber dormancy. Unfortunately, one of the most limiting factors of S. phureja is its high susceptibility to viral infections. Due to lack effective adequate diagnostic tools, infected tubers from the previous crops are frequently used as seeds which will succesively acumulate higher different types of virus resulting in a dramatic decrease in yields after several cycles. Integrated management programs of viral diseases must be supported by a good knowledge of the virus species present (and their variants) and the availability of specific and highly sensitive diagnostic tools. Currently, there is very little information on the viruses infecting S. phureja in Colombia and other andean countries. So far, there are reports of infection with Potato virus Y (PVY), Potato virus X (PVX), Potato virus S (PVS), Potato leafroll virus (PLVR), Potato yellow vein virus (PYVV) and Potato virus V (PVV). To stablish an accurate and reliable seed certification program for S. phureja in Colombia, it is paramount to perform a thorough characterization of local virus strains infecting this promising crop. This will allow the design of appropriate molecular tests such as RT-PCR and qRT-PCR that will facilitate early detection of viruses in tubers used as seed avoiding their disemination and, hopefully, resulting in crops with higher yields. This projects aims at increasing the current knowledge on the viruses infecting Solanum phureja cv. criolla by sequencing the RNA-virome using NGS techniques. This information will be useful for the design of specific molecular detection tools and will serve as basis for future seed certification programs.
