DETERMINATION OF GENETIC POLYMORPHISM TaqIA (ANKK1) TaqIB (DRD2), -141C Ins / Del (DRD2) and 40 bp VNTR (DAT1) IN COLOMBIAN POPULATION AND EVALUATION OF THEIR ASSOCIATION WITH ALCOHOLISM
Resumen
--Purpose This study aimed to analyze the polymorphisms of genes ANKK1 (TaqI A), DRD2 (TaqI B and -141C Ins / Del) and DAT1 (VNTR 40 bp) in a Colombian population of cases (alcohol dependent individuals) and controls (occasional use of alcohol) and determine its association with alcoholism. Methods 141C Ins / Del (DRD2) and 40 bp VNTR (DAT1) to a population of 214 individuals with non-problematic alcohol and individuals with problematic alcohol by PCR-RFLP. The DNA was obtained from peripheral blood white cells. Results Several hypotheses suggest that people with problem drinking have decreased density and presynaptic dopamine reuptake problems due to disturbances caused by polymorphisms associated with ANKK1, DRD2 and DAT1 genes. Gene and allelic frequencies of 214 individuals and found significant differences for allele -141C Ins in individuals who had an AUDIT score of 14 to 19 and Taq IB allele in women with problematic alcohol consumption were calculated. Conclusion The possible relationship of polymorphisms with alcohol-dependency I evaluated was obtained for the group with an AUDIT score between 14 and 19 allele frequency -141C Ins throw significant when compared with the control group differences. The present study suggests that Ins -141C allele is critical dependent alcohol consumption harmful phase. Similarly, a significant difference for allele IB TaqIB DRD2 gene polymorphism in the group of women with alcohol dependence was found. The answers to that applied to the participants in this study questionnaire made ¿¿clear that alcohol-dependent individuals are also more likely to use other psychoactive substances. Similarly there is a greater number of individuals who claim to have a family with problematic alcohol consumption in the group with problem drinking than in the control group, suggesting the influence of genetic disease.
